If I use an end mill cutter with a uniform shank, will it damage a spring collet system that uses a non-pullout button on the inside diameter of the collet?

Yes,it will likely damage the locking element and the internal taper of the collet holder. The anti-pullout element can usually be removed from the collet allowing uniform shank tools to be gripped.
